The research focuses in our team are mainly in the neutrophil and dendritic cell biology in human diseases. We have been investigating the impact of the dysregulated immune responses in terms of the overzealous neutrophil activation and type I IFN signaling cascade in the autoimmune disease and infectious diseases. The innate immune checkpoint, NLRP12 currently gains immense attention due its novel biological functions in regulating the immune resilience and maintaining the immune homeostasis. We have claimed that NLRP12 is involved in the pathogenesis of lupus nephritis in an IFN-I dependent manner. This protein is a member of NLR superfamily but is with the opposite role in the inflammasome activation as NLRP3. We are investigating the molecular mechanisms of NLRP12 in modulating the neutrophil activation in an IFN signaling dependent manner and its impact in the infectious diseases and autoimmune diseases. In addition, we also focus on a Syk coupled C-type lectin receptors (Syk-CLRs), CLEC5A which is a protective host factor in the anti-bacterial infection. Its novel roles in the activation of cytotoxic T cell and differentiation of γδ T 17 cell driven by the cross antigen presentation in dendritic cell bring about the potential application in the manipulating the host immunity in the aspect of anti-tumor immunity and immune senescence.
